Mamukkoya was the first recipient of the Kerala State Award for Best Comedian in Malayalam cinema. He began his film career with Anyarude Bhoomi in 1979 and became recognized for his distinctive use of the Mappila dialect. Over his extensive career, he appeared in more than 450 Malayalam films, including notable works such as Nadodikkattu (1987), Perumazhakkalam (2004), and Ustad Hotel (2012). His performance in Perumazhakkalam earned him the Kerala State Award for Second Best Actor. Mamukkoya also played significant roles in films like Minnal Murali (2021) and Sufiyum Sujatayum (2020).
